In one day is not enough. Might barely even be worth it unless you're able to get reservations for the Uffizzi and the academia gallery to fit perfectly into your time frame. But you won't have time to enjoy it.

We spent 2 days and it was jam packed but we felt like we got a good primer of everything it had to offer. Would definitely like to go back and spend 3-4 days next time.
